    孕酮是由黃體分泌的一種類固醇激素,食品中存在的孕酮對人體內(nèi)分泌等健康產(chǎn)生嚴(yán)重影響,。因此,,探索簡單、快速,、靈敏的孕酮檢測技術(shù)及方法很有必要,。基于孕酮的特異性適配體(Apt)功能化的Au@Fe3O4納米粒子構(gòu)建磁弛豫傳感器,,在優(yōu)化各種實驗條件的基礎(chǔ)上,,對傳感器的靈敏度,、特異性進(jìn)行評估,并對牛奶中孕酮檢測的可行性進(jìn)行分析,。當(dāng)孕酮適配體與Au@Fe3O4納米粒子的體積比為3且孵育1.5h時,,有利于Apt-Au@Fe3O4納米探針的構(gòu)建。磁弛豫檢測過程中的優(yōu)化條件如下:NaCl濃度為0.02mol/L,,Apt-Au@Fe3O4納米粒子稀釋400倍,,反應(yīng)15min。在此條件下,,水體系中單組份橫向弛豫時間的變化量與孕酮濃度間呈良好的線性關(guān)系,,線性區(qū)間為1~40nmol/L,檢測限為4.97nmol/L,,且具有良好的特異性,。該方法可實現(xiàn)對牛奶樣品中孕酮的檢測,對牛奶中孕酮的線性檢測區(qū)間為20~100nmol/L,,檢測限為13.60nmol/L,,加標(biāo)回收率為99.21%~103.45%,相對標(biāo)準(zhǔn)偏差不大于9.02%,。

    Abstract:

    With the increase of public attention on food safety, it is important to establish a sensitive and rapid detection method for progesterone (pregn-4-ene-3,20-dione, P4) in animal food products. A magnetic relaxation switch (MRS) sensor based on aptamer functionalized Au@Fe3O4 nanoparticles (NPs) was developed for the detection of progesterone in milk. The test conditions were optimized to improve the sensitivity and stability of the method, and the feasibility of this method for the detection of progesterone in milk was also analyzed.The ratio of aptamers (Apt) to Au@Fe3O4 NPs, the incubation time of aptamers modification, the concentration of NaCl, the dilute ratio of Apt-Au@Fe3O4 and reaction time were optimized to be 3, 1.5 h, 0.02 mol/L, 400 and 15min, respectively. Under the optimized conditions, the change of mono-component relaxation time (ΔT2w) showed a linear range from 1nmol/L to 40nmol/L and the limitation of detection (LOD) was 4.97 nmol/L. In addition, the proposed method also illustrated an excellent specificity. The method was also applied successfully for the detection of progesterone in milk samples. The MRS displayed linear ranges of detecting progesterone from 20 nmol/L to 100 nmol/L and LOD of 13.60 nmol/L in milk. The average recoveries of progesterone in milk were 99.21%~103.45%, with relative standard deviations of nomore than 9.02%. The results indicated that this sensor could be a potential tool for the rapid detection of progesterone in food.
