    以水體中氨氮濃度為研究對(duì)象,,在水楊酸分光光度法基礎(chǔ)上結(jié)合微流控技術(shù)進(jìn)行比色傳感實(shí)驗(yàn),實(shí)現(xiàn)對(duì)溶液中氨氮濃度的定量檢測(cè),。通過比較4種特征波長篩選方法對(duì)建模的影響,,發(fā)現(xiàn)連續(xù)投影算法選出的波長變量最少,所建立的預(yù)測(cè)模型效果最好,,建模集校正標(biāo)準(zhǔn)差和預(yù)測(cè)集校正標(biāo)準(zhǔn)差分別為0.00931mg/L和0.02857mg/L,,相對(duì)分析誤差為11.2141。在優(yōu)化條件下測(cè)得該方法的線性范圍和檢測(cè)限分別為0.04~0.92mg/L和0.016mg/L,。對(duì)養(yǎng)殖水體和海水進(jìn)行加標(biāo)回收實(shí)驗(yàn),,平均回收率在95.8%~116%之間,相對(duì)標(biāo)準(zhǔn)偏差在2.1%~6.3%之間,。結(jié)果證明了利用水楊酸分光光度法結(jié)合微流控技術(shù)檢測(cè)氨氮濃度方法的可行性,。

    Abstract:

    In order to realize the rapid and accurate determination of the ammonia nitrogen content in the aquaculture water, the ammonia nitrogen in the water was taken as the research object, and the colorimetric sensing experiment was carried out on the basis of the salicylic acid spectrophotometry combined with the microfluidic technology to realize the quantification of the ammonia nitrogen content in the solution detection. By comparing the influence of four characteristic wavelength screening methods on modeling, it was found that the continuous projection algorithm selected the least wavelength variables, and the established prediction model had the best effect. The modeling set calibration standard deviation (RMSEC) and the prediction set calibration standard deviation (RMSEP) were 0.00931mg/L and 0.02857mg/L, respectively, and the relative analytical error (RPD) was 11.2141. The effects of pH value, salinity, temperature and reagent stability were explored, and it was found that the absorbance was increased first and then decreased with the increase of pH value. The salinity had a significant effect, and temperature can speed up the reaction, and the reagents can be stored for 14 days at 6℃ in dark. Under optimized conditions, the linear range and detection limit of the method were 0.04~0.92mg/L and 0.016mg/L, respectively. The standard recovery experiment was carried out on aquaculture water and seawater. The average recovery rate was between 95.8% and 116%, and the relative standard deviation was between 2.1% and 6.3%. The results proved the feasibility of the method of using salicylic acid spectrophotometry combined with microfluidic technology to detect ammonia nitrogen.
