    摘要:

    為構(gòu)建穩(wěn)定的重組油體乳液,研究了不同超聲功率(200,、400W)和不同超聲時(shí)間(6,、12,、24min)對(duì)制備重組油體乳液穩(wěn)定性的影響,。為揭示不同超聲條件與重組油體乳液穩(wěn)定性的關(guān)系,,采用動(dòng)態(tài)激光散射,、激光共聚焦顯微鏡和圓二色譜,,研究了不同超聲條件制備的重組油體乳液流體動(dòng)力學(xué)半徑、微觀結(jié)構(gòu)和Oleosin蛋白的二級(jí)結(jié)構(gòu)變化,。結(jié)果表明,,利用超聲處理制備的重組油體乳液乳化性,、儲(chǔ)藏穩(wěn)定性和界面吸附能力遠(yuǎn)高于未經(jīng)超聲處理的乳液,并且隨著超聲時(shí)間的延長(zhǎng),,乳液粒徑逐漸變小,,乳滴形狀規(guī)則、分布均勻,,乳液表面負(fù)電荷增加,;當(dāng)超聲條件為200W、24min時(shí),,制備的重組油體乳液最為穩(wěn)定,,其乳化活性指數(shù)和乳化穩(wěn)定性指數(shù)分別高達(dá)652.2m2/g和605.2min;不同超聲處理改變了Oleosin蛋白的二級(jí)結(jié)構(gòu),,影響其與磷脂酰膽堿的結(jié)合能力,,進(jìn)而改變?nèi)橐涸谟?水界面吸附特性,,證明適宜強(qiáng)度的超聲處理有利于重組油體乳液的形成,。

    Abstract:

    Effects of ultrasonic treatments with different powers (200W, 400W) for different durations (6min, 12min and 24min) on the stability of recombinant oil emulsions were investigated. Functional properties of the composite system as a function of ultrasonic conditions were explored at the same time. Circular dichroism spectrum, particle size distribution, ζ-potential, FI and emulsibility were determined. The emulsifying properties of the reconstituted oil body emulsion was significantly increased. At the same time, the volume average particle size was decreased from 1717nm to 337.5nm, the absolute value of ζ-potential was increased, and the α-helix content was decreased to 10.4%. And the solution had homogeneous distribution and stable properties. However, with the further increase of ultrasonic power, the surface protein of the recombinant oil body was insoluble, and the functional properties of the recombinant emulsion was decreased. This indicated that the ultrasonic treatment would affect the stability of the recombinant emulsion, and the ultrasonic treatment of suitable strength was beneficial to the formation of the recombinant oil emulsion.
